2. Methodological information
Bead position measurements were taken for 2 hours a constant pulling force of 25 pN at 24C at a camera acquisition rate of 50 Hz.
Traces were cropped from the start until the end of the change in z-position
For the conversion from bead z-position to synthesized nucleotides we used the formula: Nt (F) = N * ((L(F)-Lds(F))/(Lss(F)-Lds(F))) 
F=force; N number of nucleotides in the template (2820 nt); L(F)=measured extension of the tether; Lds(F) is the full dsRNA length, and Lss(F) is the full length of a ssRNA tether
